There can be a transmission "clunk" as the clutch engages and the splines on the half shaft rotate slightly in the drive flange. If you remove the rubber cap you can see if there is grease lubricating the half shaft, mechanics who know Defenders will grease at every service thus prolonging their life.
Welding the shaft to the flange creates a one piece half shaft eliminating another transmission wear point.

Mine has recently developed a lovely clunk from the back.

If I try and drive with the handbrake on it doesn't clunk, but if I let the handbrake off and stand on the normal brakes it clunks horribly, once every time you take up drive after changing from reverse to forwards and back. It's almost like someone's strapped a cow bell underneath.

Fitting the single piece rear drive shafts to my 110 made a huge difference to backlash and how it drives.

Quick update

After 1k miles of motorway, B road and unpaved road driving the Defender feels tighter and smoother.

I've never driven a Land Rover (owned 2xSeries, RRC, 110 300tdi) with virtually no backlash before. even this 2011 110USW that had 40k on the clock when I bought it, and it's a revelation!

At very low speed on unpaved roads in the Dales it was more comfortable and generally easier to control through technical sections.

On the motorway it seemed more planted and smoother.

Another culprit of backlash is a faulty clutch switch.

I meant to mention this before but it certainly does make driving though gear changes not nice at all and does induce a backlash where if driven properly is not needed and is a nicer drive without.

You can safely bridge the clutch switch on the 2.4. un-tested on the 2.2 so cannot comment.

It is supposed to be there to reduce shock loading on the driveline, but in my option causing the backlash and delay during gear changing it will cause more wear than any shock loading would induce whilst driven properly.

Bridging it also retains all anti stall features but in low range has the benefit of reducing the antistall rev threshold down to RPM you would expect in 1st high range without the higher increased revs which is pretty annoying in low range.
Much like a rev hange in low range 1st, removed with the bridges clutch switch.

Plug and play if you use the correct socket. Diesels Lives Matter. ⛽️🛢️👨‍🔧🧰⚙️
